Main Emphasis

Within the Core Unit, we carry out histological work on animal models generated in FZB projects. In addition to consulting on pre-analysis, we process tissue for embedding in paraffin, preparation of sections, histological staining, immunohistochemistry and archiving of block material. Frozen sections are also part of our tasks. In the field of archieving, we work closely with BMB Nord. We consult the various research groups on the planning and execution of histological analyzes as well as further investigations such as transcriptomics, multispectral IHC, nanostring and spatial transcriptomics.

 

Research

As part of the German Center for Lung Research (DZL) at the North site (ARCN), we are involved in third-party funded projects in the disease areas lung cancer, COPD, asthma/allergy and biobanking. Our particular research focus is the TGF-beta signaling pathway in NSCLC. Moreover, embryological processes and the tumor microenvironment are gaining increasing relevance. After years of basic research, we are now intensively engaged in translating the results obtained in this field into clinical practice. In the area of infection, we are particularly interested in the multispectral immunohistochemical analysis of pathophysiological events in tuberculosis granulomas and transcriptome studies within various model systems and cohorts, with the aim of developing host directed therapies.

We work primarily on histological material. Its processing (FFPE, freeze, HOPE) is our central methodical basis. In addition to conventional histological staining techniques, we frequently and regularly work with immunohistochemical staining methods.

 

  • Multispectral Immunhistochemistry and Spatial Transcriptomics
    We are delighted to have a multispectral immunohistochemical platform (Akoya Vectra-Polaris) available through funding from the DZL, which, in addition to our other technologies (transcriptome, nanostring, cell culture), decisively expands the methodological portfolio. In addition, we managed to acquire a platform for spatial transcriptomics (10x) together with the research group early childhood asthma.
    We are very delighted to have a multispectral immunohistochemical platform (Akoya Vectra-Polaris) through a DZL funding, which decisively expands the methodological portfolio alongside our other technologies (transcriptome, nanostring, cell culture). In addition, we have succeeded in acquiring a platform for Spatial Transcriptomics (10x) together with the reseach group early childhood asthma.
  • Transcriptome analyses
    In addition to transcriptomics on our DZL-financed Agilent platform (incl. Bioanalyzer and Nanodrop), we offer investigations on the Nanostring N-Counter, providing a prep station. We also apply a light cycler.
  • Cell culture
    We use different cell systems to enable functional studies. In addition to cell lines, this also includes human primary cell cultures (in cooperation with the Grosshansdorf Lung Clinic). Appropriate read out systems are western blot, in cell western and fluorescence microscopy.
